Start by laying the shirt flat and identifying the exact spot where the stain is located. Now, use a paper towel to blot the area in order to absorb excess liquid. Carefully dab the paper towel in circles on the surface of the stain working your way outward.
If you happen to have a spot removal treatment formulated for fabrics such as your polo shirt, now would be time to take advantage of it. If not, you can make a simple solution of white vinegar and water. Mix one part vinegar and two parts water and apply it directly to the stained area. This will help to eliminate any bacteria that may be present. Let the vinegar solution sit on the stain for several minutes before proceeding.
Now that the shirt has been adequately prepped and treated for the stain, you can place the shirt into the wash. Set the machine for a gentle cycle and use a detergent made for delicate fabrics such as this polo shirt. Once done, hang the shirt to air dry.
If the stain persists, repeat steps 1 - 3 until you have achieved the desired results. For stubborn stains, a stronger enzyme-based detergent may be required. However, if you don't want to risk damaging the shirt, it may be better to take it to a dry cleaner.
